| Research Abstract |
1. 昨年度の実験により、ウナギの心房に貯蔵されている心房性ナトリウム利尿ペプチド(ANP)は心室に貯蔵されている心室性ナトリウム利尿ペプチド(VNP)の5倍に達するにもかかわらず、血液中のホルモンレベルは逆に1/3であった。また、金コロイド法で観察すると、心房筋細胞にはANPを含む分泌顆粒が多数存在するが、心室筋細胞中にはVNPを含む分泌顆粒が少ししか存在しないことがわかった。従って、ANPはおもに心房でつくられ、顆粒中に貯蔵されているが、VNPは心室でつくられた後に、顆粒中に貯えられることなく分泌されているという仮説を立てた。
2. ウナギの心房、及び心室の組織片培養をおこない、組織中に貯えられているホルモン量と分泌される量を比べたところ、心房ではANPの多くは組織片中に存在するが、心室ではVNPの多くは培養液中に分泌されていることがわかった。さらに、 ^<35>Sでラベルしたシスチンをもちいてパルスチェイス実験を行ない、ラベルされたホルモンの分泌速度を調べている。組織片から分泌されるホルモンの多くはプロホルモンと考えられるが、それを確認するために現在HPLC上でのANPとVNPプロホルモンの位置を調べている。
3. 心房におけるANPのmRNAの発現と、心室におけるVNPのmRNAの発現を調べている。ノ-ザンハイブリダイゼイションに用いるVNPのcDNAのクロ-ニングは既に成功したが、ANPのcDNAのクロ-ニングは現在進行中である。mRNAの発現から合成速度を予測し、貯蔵量との違いから分泌速度を推定する予定である。
