Research Abstract	ブタの白血球のアラキドン酸12ーリポキシゲナ-ゼを、モノクロ-ン抗体を用いた免疫親和性クロマトグラフィ-によって精製した。精製酵素をリジルエンドペプチダ-ゼで消化した後に、生じたペプチド断片およびN末端のアミノ酸配列を、気相ペプチドシ-クエンサ-で決定した。このペプチドの部分アミノ酸配列に基づいて2本のオリゴヌクレオチドを合成し、これをプロ-ブとしてブタ白血球のcDNAライブラリ-から酵素のcDNAをクロ-ニングした。酵素のcDNAの翻訳領域は663個のアミノ酸をコ-ドしており、分子量は74,911と計算された。ブタの各組織から抽出したRNAをアガロ-ス電気泳動で分離した後に、12ーリポシゲナ-ゼのcDNAをプロ-ブとして、ノ-ザンブロティングを行なった。酵素のmRNAは白血球に最も多く、その他に下垂体、肺、小腸や脾臓にも認められ、酵素が生体内に広く分布していることが示された。この12ーリポキシゲナ-ゼのcDNAを大腸菌で発現するために、cDNAをtacプロモ-タをもつpkkプラスミドに組み込み、発現ベクタ-を構築した。この発現ベクタ-で大腸菌を形質転換し、イソプロピルチオーβーDーガラクトシドで酵素タンパクを発現誘導した。この大腸菌の抽出物をアラキドン酸と反応させ、反応産物を高速液体クロマトグラフィ-で分析すると、12ーヒドロキシ酸と同定され、大腸菌で発現された蛋白が12ーリポキシゲナ-ゼ活性をもつことが示された。さらに、酵素に対する抗体を用いたウェスタンブロティングによって大腸菌の抽出物で陽性バンドが認められ、大腸菌で酵素蛋白が発現していることが確認された。
