| Research Abstract |
I.プロ型前駆体変換酵素遺伝子のクロ-ニングと一次構造の解析.
1)ヒトおよびラット肝cDNAライブラリ-からそれぞれ4.7kbおよび4.3kbのFurinに対するcDNAクロ-ンを得た。塩基配列から推定されるFurinの一次構造は,ヒトで794残基,ラットで793残基のアミノ酸から構成されており,N末端側に約300残基のズブチリシン類似の構造およびC末端近くに膜貫通部位に相当する配列をもつことがわかった。ヒトとラットの間には93.7%の相同性がみられた。
2)アルブミンおよび補体C3のcDNAをトランスフェクトしたCOSー1細胞では,合成された各プロ型前駆体の約50%はプロセスされずに分泌された。これにFurinを発現させると,各プロ型前駆体は完全にプロセスされることがわかった。本研究で得たFurinはプロ型前駆体変換活性をもつエンドプロテア-ゼであると結論される。
II.プロ型前駆体変換酵素に対する特異的インヒビタ-の調製と応用。
1)ラット肝cDNAライブラリ-からα_1ープロテア-ゼインヒビタ-(α_1ーPI)およびコントラプシンのcDNAクロ-ンを得た。部位特異的変異手法により,α_1ーPIの352番目のMetをArgに,またコントラプシンの359番目のLysをArg変えて,プロテア-ゼとの反応部位にArgーSerをもつ変異体をコ-ドするcDNAを調製した。
2)COSー1細胞におけるトランフェクション実験の結果,発現された各変異型インヒビタ-はプロアルブミンおよびプロC3のプロセシングを完全に阻害することがわかった。正常のα_1ーPIやコントラプシンおよび反応部位に同じArgーSerをもつアンチトロンビンIIIにはこの阻害活性が認められないことから,本研究で得た変異型インヒビタ-はプロ型前駆体変換酵素に特異的なインヒビタ-として利用できることがわかった。
