肺リン脂質代謝調節におけるサ-ファクタント・アポ蛋白関与の機構

Research Abstract

本研究は、肺胞II型細胞におけるリン脂悉質代謝調節へのサ-ファクタント・アポ蛋白,特にSPーAとSPーD関与の機構を解明することを目的とする。前年度の研究成果に基づき立案された平成3年度の研究計画は,概ね達成された。平成3年度の研究実績を以下に要約する。
1.主要アポ蛋白のSPーAは,サ-ファクタント機能を担うリン脂質,ジパルミトイル・ホスファチジルコリン(DPPC)に特異的に強く結合することを発見した。DPPCがSPーAに特異的に結合することの代謝的意義が次に検討された。DPPCを含むリポソ-ムが,不飽和PCを含むリポソ-ムよりも,肺胞II型細胞より分離した形質膜に,SPーAの存在下で撰択的に結合した。またDPPCを含むリポソ-ムは,肺胞II型細胞へSPーA存在下に撰択的にとり込まれた。細胞内へとり込まれたDPPCはラメラ封入体へ蓄積した。すなわち,SPーAは肺胞II型細胞におけるDPPCの再利用を撰択的に促進することが示された。
2.Ca^<2+>存在下でのリン脂質リポソ-ムの凝集及び肺胞II型細胞へのリポソ-ムの取り込み作用発現には,SPーAの糖鎖部位は重要でなく,コラ-ゲン様構造が必須であることを示した。SPーAの機能発現には高次構造の維持とCa^<2+>の構造が必要とされるが,ジスルフィドに結合の切断はSPーAへのCa^<2+>結合数に影響しなかった。
3.親水性アポ蛋白のSPーDは,主として細氣管支上皮細胞に存在することが抗SPーD抗体を用いた免疫電顕で明らかとなった。SPーDのリン脂質結合性は極めて特異的で,ホスファチジルイノシト-ル(P1)にのみ結合した。SPーDのPI特異的結合の生理的意義は不明だが,SPーDはPIの膜間移送活性はなかった。
4.周産期におけるSPーA,SPーB,SPーDの変動パタ-ンは異なり、別個に調節されているが,SPーAとDPPCの変動パタ-ンは一致した。
