| Research Abstract |
1)胃腺腫、胃癌におけるRAS、MYC、FOS、ERBB2、EGFR遺伝子の免疫組織化学的検討は終了した。胃癌におけるERBB2遺伝子の発現に関しては論文投稿中であるが、胎児、新生児胃粘膜では発現が強いこと、胃癌では印環細胞癌を除けば、組織型は関係なく高率に発現していることが明らかとなった。
胃腺腫における癌遺伝子産物については論文準備中である。
2)パラフィン包理ブロックよりのDNA抽出は2/3程度の症例で可能であった。slot blot法による検討では胃腺腫ではMYC、FOS、ERBB2遺伝子の増幅を認めなかった。Polymerase chain reaction(PCR)法を用いPAS遺伝子の点突然変異についてみたところ、胃腺腫24例中1例でのみ点突然変異が見い出された。
3)TGFβ免疫活性は癌細胞のみ陽性であったがsialidase処理(2時間)にて正常粘膜上皮にも陽性となった。これは不活性型TGFβが活性型になった為と見做された。
4)p53免疫活性は胃癌の約50%で陽性癌細胞の出現を認めた。この結果はWestern blot法による検討とよく一致していた。地方、パラフィン包理切片を用いた検討では胃腺腫10例いずれにも陽性細胞を見い出さなかった。
5)胃癌の新鮮凍結材料よりDNAを抽出し、RFLPマ-カ-を用いた遺伝子欠失の検出を開始した。用いるプロ-ブは8個の染色体上の14種である。
