| Research Abstract |
1.ヒトおよびブタ各組織におけるエンドセリン(ET)受容体サブタイプの解析:ヒトおよびブタ各組織より調製した膜標品を用いてのAffinity labelingと結合実験の結果,各組織には主として2つの型のET容体,即ちETー1,ETー2に親和性を有するETー1/ETー2特異受容体(ET_A)とET isopeptide全てに親和性を有するET family共通受容体(ET_B)が部位特異的に分布していることを見い出した.即ち,心血管系では主としてET^A受容体、脳、副腎、腎では主としてET_B受容体、肺では両者が分布していた。また、血管系では内皮上にET_B受容体、平滑筋上にET_A受容体が局在、各々、弛緩と収縮に連関しており,血管でのET受容体サブタイプの機能分化を証明した。
2.エンドセリン(サラホトキシン)アフィニティ-クロマトグラフィ-の開発:種々のET及びET類似ペプチドのサラホトキシン(SRT)の誘導体を合成、SRTの4位のリジンをbiocytinと置換したペプチド、[Bio^4]SRTが最適リガンドであることを見いだし、アビジンを介してアガロ-スに架橋し、[Bio^4]SRTカラムを作成、可溶化受容体の結合、解離に成功した。
3.ウシ脳のET_B受容体の精製し一部アミノ酸構造の決定:[Bio^4]SRTカラムとさらにゲル濾過によりウシ脳よりET_B受容体蛋白を得た。分子量は50Kであった。プロムシアン断片ペプチドより一部アミノ酸構造を決定した。
4.ヒトET_B受容体cDNAのクロ-ニングによる一次構造の決定:ウシ脳と肺からの精製ET_B受容体の一部アミノ酸構造を基にオリゴヌクレオチドプロ-ブを合成,ヒト肝のcDNA libraryよりET_B受容体をクロ-ニング、その一次構造を決定、さらにCOS細胞で発現させ、結合特異性を検討した。さらに、他の受容体サブタイプのクロ-ニングを試みている。
