Research Abstract |
By the immunocytochemical, immunochemical, and immunoelectron microscopic method using monoclonal antibodies against predictive synthetic beta-protein with the sequence of pesidure 597-620, the distribution and localization of beta-ainyloid protein precursor(beta-APP)in cultured vascular cells was investigated. beta-APP immunoreactivity was demonstrated in the plasma membranes of the cultured vascular cells as well as in the cytoplasma of the cells. Cytoplasmic beta-APP immunoreactivity located in the proximity of the cell nucleus and looked as microbody structure with various size. beta-APP immunoreactivity was also demonstrated on the surface of the basement membrane formed in the plastic dish during the culture of the cells. These findings suggest that the abnormal production or secretion of beta-APP in the vascular wall cells could be a cause of the formation of amyloid angiopathy in the Alzheimer's disease.
