末梢神経障害における神経成長因子の遺伝子発現とその調節機構に関する研究

Research Abstract	平成2、3年度に引き続き末梢神経障害の変性再生過程における神経栄養因子およびその関連分子の再現動態を検討した。本年度は以下のような知見が明らかとなった。1)培養シュワン細胞にCAMPを作用させるとNGFmRNAは増加、LNGFRmRNA、BDNFmRNAは減少の方向に変化した。さらにlaminin B_1、B_2 mRNAは増加に、typeI、III collagen mRNAは減少する方向に変動した。これらはシュワン細胞のこれらの分子の産生調節にCAMPが重要であることを示している。2)ヒトの剖検例についてLNGFR,trk,trkBの末梢神経系における発現分布を検討したところ、LNGFRmRNAは広範な発現がみられたがtrkは交感神経、感覚神経細胞体に限局して発現されていた。trkBについては脊髄、末梢神経にも発現がみられた。3)ヒトおよびラットの末梢神経についてLNGERとtrKの発現の加齢変化を検討したが、高齢に至ってもこれらの発現はよく維持されていた。4)培養交感神経感覚神経に対するNGFの作用を検討したがこの両者でneuriteの分岐に対する反応性およびその加齢変化が異なることが明らかとなった。以上の所見は末梢神経障害におけるこれらの分子の発現動態を明らかにするものと考えられるが、今後更にこれらの分子の発現調節に係わる因子の解明や実際のヒト末梢神経障害における役割などについて明らかにして行く必要があると考える。
