新しい基底膜浸潤モデルを用いた悪性腫瘍の転移の分子機構に関する研究

Research Abstract

我々が1986年に開発した、再構成基底膜を用いたin vitro invasionassayは、今や世界で最もポピュラーな腫瘍細胞の浸潤能の測定法となっている。しかし、従来、このアツセイを用いた研究結果は、すべて樹立された細胞株に関するものであった。もしも、外科手術や生検でえられた腫瘍についてのアツセイが可能となれば、個患者での転移能の予測、生命的予後の判定因子として有力なアツセイ法となる可能性がある。我々は、外科手術や生検でえられた腫瘍のprimary cultureでこのinvasion assayが可能なことを発見した。骨肉腫、Ewing肉腫、悪性神経上皮腫では強い浸潤能を示し、脂肪肉腫では以上より多少弱いが、やはり浸潤能を示した。しかし、正常の皮フ組識よりえられた線維芽細能は浸潤能を全く示さなかった。今後さらにデータを集積し、長期的に患者の肺転移、生命予後の観察を行ない、invasion assayが臨床における予後判定法として用いることができるかどうか判定したい。
腫瘍細胞を、再構成基府膜(matrigel)に浮遊させてタードマウスの皮下に移植すると、タードマウスへの可移植率が促進されることを発見し報告した。まず、HT1080ヒト線維肉腫細胞あるいはMG63ヒト骨肉腫細胞という樹とされた細胞株につき、matrigelで可移植率が促進されることを確認した。次に、悪性骨軟部腫瘍患者の手術材料、生検材料についても、同様に、可移植率が促進されることを確認した。従来、ヒト悪性腫瘍のヌードマウスへの移植の問題点は、可移植率が40-50%であり、被検材料や高価なヌードマウスを無駄にしやすいという点にあった。今回の、matrigelにより可移植率が上がるという発見は、制癌剤感点性テストなど、ヌードマウスを用いた腫瘍研究の福音になると思われる。
