Research Abstract	Ig遺伝子プロモ-タ-に結合するOCT2たん白をコ-ドする遺伝子pucC_1は免疫グロブリン産生を促すことが知られており,それ故免疫グロブリン産生の障害を示す免疫不全症の成立に何らかの役割を演じていると考えられる。そこで我々はOCT2たん白に対する抗体を作製することを目的にOCT2遺伝子を発現ベクタ-に挿入し,in vitroでたん白を翻訳させることを考えた。IgGFc部分との融合たん白として本たん白を回収すべくPRIT2Tベクタ-にOCT2遺伝子をin frameに結合させ(そのためまずBluescriptベクタ-に本遺伝子を結合させ,その後再びそれを切り出しrestriction siteを変化させPRIT2Tベクタ-に結合させた。),大腸菌にとりこませたのち,IgGセファデックスカラムでたん白の回収に努めた。併し本たん白がDNA結合たん白であるせいかOCT2たん白の回収はできなかった。そこで作戦を変更し、本遺伝子をpGeーmini1ベクタ-に結合し,T7プロモ-タによってin vitroでmRNAを合成させることにした。その結果予想されるサイズよりやや小さめであるがエチジンブロマイド染色によって認められる程度のmRNAを得ることができた。現在更にRabbit Reticulocyte Lysateの系を利用してin vitro translationの実験に移っている。更にReverse Transcriptaseを用いたRTーPCR法によってOCT2mRNAの発現あるいはその変異の発見等にもアプロ-チしていく予定でいる。
