bach1/bach2二重ノックアウトマウスを用いたヘム応答性遺伝子制御システムの解析

Research Abstract

この半年間に実施した主な実験は以下の通りである。
1.Bach1およびbach2ヘテロノックアウトマウスを交配し,bach1^<-/+>bach2^<-/+>マウスを得た。現在,これをさらに交配することにより,二重ノックアウトマウスの作出を試みている。
2.組み換えBach1を大腸菌から精製しウサギに免疫した。そしてBach1特異的抗体を得ることに成功した。現在,内在性Bach1の検出に使えるかどうか,ウエスタン法,免役沈降法,クロマチン沈降法を用いて検討中である。
3.heme oxygenase-1 (HO-1)遺伝子座のクロマチン構造を検討するシステムを構築した。すなわち,DNase I感受性を用いてクロマチンの凝集状態を調べることが可能になった。また,アセチル化ヒストンとメチル化ヒストンに対する抗体を用いたクロマチン沈降法により,HO-1誘導前後でのクロマチン修飾を比較できるようになりつつある。
