Research Abstract	創成的基礎研究「生体機能の統合的研究」班は新しい研究分野を切り拓く計画を練り、平成2年度に始まる向う5ケ年の研究計画として出発した。本研究班は高次脳機能を中心に統合的な生体機能の解明を目標として,総括班および3つの研究班,即ち,(1)脳の高次機能の非侵襲的アプロ-チ(佐々木班長),(2)行動制御による高次脳機能の研究(丹治班長),(3)高次脳機能の基礎としての神経回路の働きの研究(江橋班長,外山副班長)を編成し,研究課題への取組みを開始した。平成2年9月28日に第1回目の研究班全体会議および3つの研究班の個別研究会議を開催して新プログラムによる国際共同研究を目指す創成的基礎研究の性格の周知をはかると共に,これからの取組みの焦点となる研究課題についての討論と協議をおこなった。平成3年2月15日および16日には第2回目の全体会議および個別研究班会議を開催し,本年度に発足した創成的基礎研究の進展状況を把握すると共に,個々の研究課題についての研究報告と次年度以降の展望を討議した。研究総括班は本研究の全体の研究計画,研究推進の方策,共同研究のための比較的高額な研究機器の配置,国外からの研究分担者の招へい,研究成果の取り纏めと評価を担当することとした。研究第1班は脳の高次機能の非侵襲的アプロ-チへの取組みを開始した。脳磁計を用いた研究ではヒトの感覚機能・認知機能に関する新知見,小動物を用いた磁界発生点の検証等が報告され,生理学研究所に新たに設置された脳磁計(MEG)による研究も端緒についた。ヒトの感覚,運動系の脳電図や行動解析による脳機能へのアプロ-チも進展している。磁気共鳴装置(MRI)による脳の三次元イメ-ジへの試みも開始された。研究第2班は高等動物の多様な運動と行動を制御する機構を脳の多くの部位にわたって探索し,神経細胞活動の基礎との関連に及ぶ多彩な研究が開始された。例えば,霊長類大脳皮質のなかの,前頭前野・運動前野・運動野の機能的関連の研究,眼球運動制御を行う大脳基底核の役割,特に黒質と尾状核のニュ-ロン活動に基く眼球運動の状態の研究,大脳運動領野が広範な拡がりを示すとともに小領域への細分化を示唆する知見,その根拠となるニュ-ロン活動の特性の研究等が進められた。研究第3班はさまざまなin vitro標本を用い,神経回路の生後発達の先天性および後天性機構に関する研究を行った。1)視覚系のcoーculture標本により,視覚野のIIーIII層から対側の視覚野,V層から四丘上丘,VI層から外側膝状体への遠心性投射,外側膝状体からIV層への求心性投射に関する層特異性は全て先天的機構に依存して形成されることが示された。2)また小脳系のcoーculture標本ではプルキンエ細胞のシナプス学習に長期増強と長期抑圧の2つの機構が含まれ,前者は顆粒細胞の単独刺激で生じ,後者は顆粒細胞と下オリ-ブ核刺激の同時刺激で生じることなどが示されるなど大脳あるいは小脳皮質系の学習の先天的および後天的神経機構について大きな研究成果を得た。
