Research Abstract

遺伝性多飲マウスの飲水調節機構の分子・生理学的研究
遺伝性多飲マウスの飲水調節機構について調べる。まず免疫組織化学法により脳内のバゾプレッシン、オキシトシン、VIP(vasoactive intestinal polypeptide)およびcーfos蛋白について調べた。正常マウスに比べて遺伝性多飲マウスの方が視床下部のバゾプレッシン陽性細胞の数が多かった。オキシトシンとVIPについては差がなかった。cーfos蛋白は脱水負荷後に視床下部に、遺伝性多飲マウスと対照マウスとでは異なった出現パタ-ンがみられたが詳細は検討中である。また、in situ hybridization法によりバゾプレッシンのmRNAの発現を組織学的に調べた。対照マウスと比較して遺伝性多飲マウスの方が視床下部室傍核と視索上核とに多くのシグナルがあった。さらにNorthern blot分析法により視床下部のバゾプレッシンmRNA量を測定したところ、遺伝性多飲マウスでは対照マウスの4倍の量があった。そこで多飲の原因が脳内のバゾプレッシングが過剰なことによるという仮説を行動学的に検証するために以下の実験を行った。まずバゾプレッシンの抗血清1μlを側脳室に微量注入した。注入前日の飲水量は37.9±1.5ml(平均±標準誤差)で、抗血清を投与した日は20.9±4.6mlになり、有意に減少した。血清中の補体成分を失活させ、抗体成分に影響しないとされる60℃で30分の熱処理を加えた血清を投与した場合も飲水量は28.4±5.4mlに減少した。生理食塩水を投与した場合には飲水量に変化はなかった。正常マウスも通常時の一日の飲水量が10.2±1.6mlであるのに対してパゾプレッシンの抗血清の投与によって4.8±1.0mlに減少した。次にバゾプレッシンの桔抗薬である[Pmp^1,Tyr(Me)^2]ーArg^8ーVasopressin3ー300ngを同様に投与した。用量によらず、飲水量は投与後24時間で10ー14mlと生理食塩水を投与した場合の34.2±5.9mlに比べて有意に減少した。以上の結果より遺伝性多飲マウスは脳内のバゾプレッシンが過剰であることが原因で多飲となっていることが示唆された。
