Research Abstract |
Varicu intractable diseases have been known to originate from metabolic disorders and abnormalities in signal transduction. Howejer, the detailed mechanisms of the causal relationship between them have not been fillly understood. The project was aimed to elucidate the mechanisms by which metabolic processes as well as signal transduction are regulated, and to promote mutual understanding in scientific research on these fields between the scientists from Japan and China. The following investigations were carried out : 1. Scientific exchange on the metabolic regulation between Japan and China : Two scientists from Jilin Medical College, China, visited Japan from Oct. 1, 1991 to Dec. 17, 1991. During this period, they engaged in collaborative works on metabolic regulation and signal transduction in Shiga University of Medical Science. Role of oxygen in the metabolic regulation : Besides the well known role of oxygen in the energy metabolism, oxygen has been shown to play an important physiological role in metabolic regulation and signal transduction in the cells through oxygenase-catalyzed reactions. we have studied the well known oxygenase, catechol dioxygenase as follows : a) An enzyme which cleaves the benzene ring of 3, 5-dichlorocatechol has been purified to homogeneity from a Pseudomonad, grown with 2, 4-dichlorophenoxyacetic acid (2, 4-D) as the sole carbon source. The enzyme was a nonheme ferric dioxygenase and catalyzed the intradiol cleavage of 3, 5-dichlorocatechol. Thus the enzyme was designated as 3, 5-dichlorocatechol 1, 2-dioxygenase. Judging from the substrate specificity, kinetic constants, and N-terminal amino acid sequence, the enzyme was found to be distinct from other chlorocatechol dioxygenases reported previously. b) Trials were made to substitute the iron cofactor in the active site of catechol 2, 3-dioxygenase by other metals, and to modify the enzyme by sulfhydryl (-SH) blocking reagents or by suicide substrates. we obtained several basic information on the enzyme. 3. Biosignal transduction and its abnormalities : The mechanism of abnormal sensation in patients with peripheral nerve disorders such as carpal tunnel syndrome or cubital tunnel syndrome remained unknown. we used the technic of microneurography to detect the relationship between subjective numbness of these patients and objective neural activities of the sensory nerve. This study showed that affected sensory nerves were spontaneously activated in patients with these diseases. In addition, microneurograms demonstrated that some-types of-myelinated large fibers of the median (ulnar) nerve are more activated when patients felt paresthesia in the hands. Less
