| Research Abstract |
CBAーlpr^<cg>/lpr^<cg>とCBAー+/+との間で相互の骨髄移植を行った。CBAーlpr^<cg>/lpr^<cg>→CBAーlpr^<cg>/lpr^<cg>ではリンパ節腫脹、自己抗体共に出現、CBAー+/+→CBAー+/+およびCBAー+/+→CBAーlpr^<cg>/lpr^<cg>では両者共に出現せず、CBAーlpr^<cg>/lpr^<cg>→CBAー+/+では自己抗体は出現しながら、リンパ節腫脹は出現しなかった。ところがこの最後の骨髄キメラの肝臓には異常リンパ球の浸潤が見られたので、それに腫脹前のCBAーlpr^<cg>/lpr^<cg>由来リンパ節を移植したら、有意な腫脹が見られた。このことより、リンパ節腫脹は骨髄より由来する異常リンパ球と異常リンパ節との相互作用により生ずることを明らかにした。そこで、lpr^<cg>、lpr、gld各々からの骨髄、リンパ節の組合せを作成し、レシピエントとして(CBA x C3H)F_1を用いてリンパ節腫脹の遺伝的規定について調べた。骨髄がlpr^<cg>/lpr^<cg>、lpr/lprどちらの場合でもlpr^<cg>/lpr^<cg>、lpr/lprのリンパ節を腫脹させたが、gld/gld、+/+のリンパ節を腫脹せさなかった。これに反してgld/gld骨髄はlpr^<cg>/lpr^<cg>、lpr/lpr、gld/gld、+/+のいずれのリンパ節をも腫脹させた。これにより、lpr^<cg>とlprとは骨髄およびリンパ節レベルにおける機能表現が同じであり、gldとは異なるものであることを明らかにした。これはlpr^<cg>とlprとのallelismを支持するものである。さらに、lpr^<cg>/lpr^<cg>骨髄を移植されたレシピエントにlpr^<cg>/+のリンパ節を移植したところ、軽度ながらも有意な腫脹を示し、ヘテロ接合lpr^<cg>はリンパ節レベルにおいてsemidominantに機能表現することを明らかにした。我々はまた、CBAーlpr^<cg>/lpr^<cg>とDDDーnu/nuとのintercrossにより、lpr^<cg>/lpr^<cg>.nu/nuの無胸腺ダブルミュ-タントマウスを作成し、それらへの胸腺移植も行い検討した結果、lpr^<cg>によるリンパ節腫脹や自己抗体産生などは胸腺依存性であること、その際の胸腺としては異常胸腺である必要はないことを明らかにした。
