Research Abstract	抗ダンシルマウスモノクロ-ナル抗体に結合したハプテンE‐DNSLysのコンホメ-ションを決定し,抗原結合部位を反応の場として用いて,ハプテンアナログ体(DNS‐levulinic acid)の不斉還元応反を行った。 TRNOE測定により巨大分子に結合している状態の低分子のコンホメ-ションを,巨大分子に結合していない低分子のスペクトルから決定する。抗体に結合した状態のDNS‐LysのTRNDE測定を行ったところ,DNS環のみならずLys側鎖にも隣接プロトンの間に負のNOEが観測された。したがって,抗体によりDNS‐Lys分子全体が認識されていることが示され,抗ダンシル抗体ではハプテン側鎖までも抗原認識において重要な役割を果していることが明らかとなった。 Lys残基の代りに側鎖にケトン基をもつlevulinic acidを用いてDNS‐levulinic acideを合成し,同様にTRNOE測定を行ったところ,DNS‐levulinic acidも,抗体に結合しているDNS‐Lysと同様のコンホメ-ションをとっていることが明らかとなった。そこで,抗体存在下でNaBH_4を用いてDNS‐levulinic acidのケトン基の不斉還元反応を行った。反応物をキラルカラムに流しS体とR体を定量したところ,抗体非存在下ではS体とR体の比率は5:5であったが,抗体存在下では7:3となり,明らかに不斉収率の向上がみられた。以上の結果は,抗体の抗原認識部位を有機化学反応の場として用いることを示している。
