Research Abstract	本研究班は「大腸菌ゲノムの全体像」の中で(ゲノムの機能的全体像)を分担しており、大腸菌の遺伝子群の機能とその発現制御系を中心に研究を進めてきた。中田と牧野はugpオペロン(snーグリセロ-ル3ーリン酸能動輸送系をコ-ド)の発現が、リン酸レギュロンの調節遺伝子であるphoR/phoBと、cAMPーCRP複合体とによる二重の調節を受け、また、ackA(アセテ-トキナ-ゼをコ-ド)の過剰発現がリン酸レギュロンの発現を誘導するなど、リン酸レギュロンが他の調節系とも関連して統合的に制御されていることを示した。饗場はcrp(cAMP受容体蛋白質をコ-ド)の転写の正の自己制御機能が働くことを証明し、菌の成育条件の変動に応じて正負双方の自己制御でcrpの転写が調節されていることを示した。安楽は嫌気性亜硝酸還元酵素(チトクロ-ムc552)の正の調節遺伝子dniRをクロ-ニングして塩基配列を決定史、チトク-ムbo複合体の活性中心の解析から電子伝達の複核中心モデルを提唱した。沓掛はゲノムデ-タベ-スから鞭毛遺伝子特異的シグマ因子(RpoR)依存性プロモ-タ-と相同性の塩基配列を7個見いだし、その1個は実際にRpoF依存的に転写されることを証明した。嶋本は一本鎖DNA結合蛋白質(SSB)の過剰産生菌の蛋白質を二次元電気泳動法で分析し、その発現量に反比例して発現する蛋白質を7種類発見した。二井はATP合成酵素の活性中心と構造、H^+輸送路およびATP合成/分解とH^+輸送の共役機構の解析を行なった。松橋は細胞の成長と分裂に関与するmra遺伝子群(遺伝子地図上2分)の上流のmraRが細胞成長と分裂に重要な役割を演じていることを証明した。由良は熱ショック応答遺伝子の転写に必須のシグマー32のmRNAが定常期では二次構造を形成して殆ど翻訳されず、熱ショックなどのストレスにより、それが一次的に解けて翻訳誘導が起こるというモデルを提唱した。
