糖鎖抗原の癌性変化とその臨床応用に関する基礎的研究

Research Abstract	本研究の目的は、癌化にともなう細胞表層の糖鎖抗原の変化とその生理的意義を解明し、ヒト癌の診断と治療への応用の基礎とすることである。我々は今年度、悪性細胞の血管外浸潤や血行性転移に関連するとみられる糖鎖抗原の細胞接着因子としての機能について検索した。シアリルSSEAー1抗原は白血球や肺癌、卵巣癌に強く発見されている。しかし、消化器系の癌細胞では、SSEAー1系の2型糖鎖よりも、2→3シアリルLe^a抗原など1型の癌関連性糖鎖抗原の方が陽性率が高い。我々は今年度、実はこの2→3シアリルLe^a抗原(1型糖鎖抗原)もELAMー1のリガンドであることを明らかにした。ヒト培養大腸癌細胞Colo201は2型のシアリルLe^x抗原を中等度に発現するが、1型の2→3シアリルLe^a抗原をより強く発現する。この細胞もまた血管内皮細胞のELAMー1によく接着するが、意外なことにこの接着は抗シアリルLe^x抗体よりむしろ抗2→3シアリルLe^a抗体で強く阻止される。これは、シアリルLe^a陰性のヒト白血病細胞HLー60の接着が抗シアリルLe^x抗体のみで特異的に阻止されるのと対照的であった。両糖鎖抗原がともにELAMー1のリガンドであれば、2→3シアリルLe^aのリポゾ-ムもシアリルLe^xのリポゾ-ムも、Colo201およびHLー60のどちらの細胞の接着をも阻止するはずである。以後の研究は愛知県がんセンタ-にて継続の予定である。
