Research Abstract	筋形成について、中胚葉誘導、細胞分化誘導、脱分化、遺伝子転写制御、収縮構造の形態形成など種々のレベルでの分子機構を検討した。 1)浅島はツメガエルの細胞培養液からアクチビンA、AB、Bおよびフォリスタチンを単離・精製し、濃度依存的な中胚葉誘導作用の詳細を解明、さらに高濃度アクチビンによる腎臓や心臓の分化誘導もを見いだした。 2)筋細胞系列の決定と分化、脱分化について、佐藤は原索動物ホヤで自律的に筋細胞分化を導く因子を探索し、UV照射により筋分化能を失った卯細胞に筋分化能を付与できる卵断片を得た。遠藤は筋分化、脱分化を可逆的に起こせる筋細胞株(SV40Tを導入したC2細胞)を用い、分化と脱分化時における細胞周期誘導因子群の発現変化を明らかにした。小沢は鶏胚の体節筋芽細胞の肢芽筋形成領域への移動に際し、ガイドとしての側板側の重要性を見いだし、それに関わる因子を検索した。 4)筋蛋白質遺伝子とその転写制御について、鍋島はミオシン軽鎖遺伝子の転写制御を解析、エンハンサーに存在するE-ボックスはMyoD、Myogeninと選別的に反応すること、プロモーター領域でもMyoDとmyogeninが異なる作用を示すこと、さらに転写制御因子群の複合作用の重要性をを明らかにした。佐藤はホヤでclusterをなす5つのアクチン遺伝子の構造を解明、その内で筋特異的に発現されるHrMA4について転写制御域を明らかにした。 5)収縮構造の形態形成に関連して、眞崎は新たに見いだされた115-kDa-αアクチニンの構造の決定と組識および発生段階依存的な発現、細胞内局在を明らかにした。大日方はアクチン集合制御因子コフィリンの筋型アイソフォームのcDNAを分離、構造と発現を解明、また筋細胞への蛍光標識コフィリンの導入およびコフィリンcDNAのトランスフェクション法により筋細胞内でのコフィリンの作用のしかたを明らかにした。嶋田は培養筋細胞にセンス、アンチセンスcDNAの導入によりトロポニンC、Iの発現量を改変、そのときの形態形成を調べた。
