Research Abstract |
筋形成の一連の過程(筋細胞系列の決定、分化誘導、筋蛋白質遺伝子発現、収縮構造形成)の分子機構について研究を進め、以下の成果をあげた。 1)中胚葉誘導因子アクチビンの濃度依存的な誘導と誘導される側の部域と発生段階特性を詳細に解明、さらにツメガエルの筋分化因子Myf5の発現制御を解析した(浅島)。 2)筋分化制御因子であるMyogenin遺伝子の欠失マウスをジーンターゲット法により作成、筋形成への影響を探り、Myogeninの欠失でも筋芽細胞は生ずるが成熟した筋細胞に至らないことを見いだした(鍋島)。 3)ホヤの5種の筋肉アクチン遺伝子クラスターおよびそれらの発現特性を解明した(佐藤)。 4)筋細胞の分化/脱分化におけるc-jun、cdk2/cyclin、cdc2/cyclin、Rbの活性化と不活性化の相関を明らかにした(遠藤)。 4)筋細胞株(C2)をマウスに注入、生体内で分化へ導く実験系をつくった(小沢)。 5)収縮蛋白質の構造への集合について。形成期の筋で蛋白質集合制御に関わるプロフィリン(大日方)、115kDa αアクニチン(真崎)、C-蛋白質(心筋型)のcDNAを分離、全構造を決定した。アクチン制御蛋白質コフィリンが筋萎縮時に高発現することを見いだした(大日方)。ビチオン標識アクチンを培養心筋細胞に注入、アクチンの筋原繊維への集合過程を電子顕微鏡で解明した(嶋田)。
