Research Abstract

本総合研究は細胞接着因子の機能シグナルに関するものである。各分担者の研究成果は概ね予定通り進展し、成果として以下のようなものが得られた。機能シグナルを化学構造から分類するとコラ-ゲンファミリ-、細胞接着性糖タンパク、プロテオグリカンおよびそれらには分類出来ないものに分けられる。コラ-ゲンの遺伝子構造解析からV型コラ-ゲンα1鎖が決まった。活性型アスコルビン酸のコラ-ゲン合成促進作用機構の解明が進んだ。再構成コラ-ゲンゲルを用いた培養細胞の研究から、収縮したゲルによる細胞増殖停止、細胞形態との関わり、さらにコラ-ゲンゲル収縮における細胞性フィブロネクチンの役割で新たな発見があった。また、コラ-ゲン結合性のフォンビルブランドプロポリペプチドその他のタンパク分子中の結合部位構造をより狭い領域に限定できた。プロコラ-ゲン結合性HSP47の機能について新し展開がなされた。糖タンパク質ではフィブロネクチン、ラミニン、テネイシン、ビトロネクチンの構造と機能に関連して、いくつか新しい知見が得られた。細胞接着ドメインとしてRGD以外の構造が存在し、RGD接着シグナルの重複性と接着シグナルの多様性が判明した。ラミニン分子の多様性と細胞特異性についての手がかりが得られた。
細胞接着の機能シグナルには多様性がある。一方、造血幹細胞の維持におけるプロテオグリカンを初めとするマトリックス成分の役割などの研究から、一般にマトリックスが細胞の増殖、分化、遺伝子発現など細胞機能の根幹に関わることが明らかになり、細胞外基質と細胞の間の関係(接着の三次元的な配置など)が細胞の極性、細胞の組織的な集団化が分化に重要であることが判明した。今後、細胞機能発現における接着因子シグナルの普遍性と特異性の解明をさらに掘り下げることを目標の一つとする。
