Research Abstract

HLAによる免疫応答性の制御機構を解明するために,溶連菌細胞壁抗原(SCW)に対する免疫応答性を解析した。昨年度は溶連菌細胞壁抗原(SCW)に対する低応答者から免疫抑性活性を有し,αβ型T細胞レセプタ-(TCR)を発現した自己反応性CD8^+T細胞株が増殖すること,さらにそのCD8^+T細胞が抗原提示細胞に対する細胞傷害活性を有することを明らかにした。今回この傷害活性が認められる条件および自己反応性CD8+T細胞のT細胞レセプタ-(TCR)と認識するHLAの同定を行った。
(1)自己反応性CD8^+T細胞の末梢血単核球,B細胞株に対する細胞傷害活性には,末梢血リンパ球培養上清中の液性因子が必要であった。通常の細胞障害性CD8^+T細胞はこのような液性因子を必要とせず、今回得られたCD8^+T細胞のユニ-クな点であった。この液性因子は、既知のリンホカインでは置き換えられなかったため。本年度は液性因子の精製に必要となる大量の培養上清を作製した。現在この液性因子の精製および構造決定を行なっている。(2)自己反応性CD8^+T細胞株のTCRを単クロ-ン抗体を用いて検討したところ、TCRVβ5.2陽性、TCRVα2陽性細胞とTCRVβ5.2陽性、TCRVα2陰性細胞からなっていた。TCRVα2陽性および、陰性細胞に分けて、CD8^+T細胞の拘束分子検討した。パネル細胞およびモノクロ-ナル抗体を用いた解析により、TCRVα2陽性CD8+7細胞は標的細胞上のHLAーBw54分子をTCRVα2陰性CD8^+T細胞はHLAーBw52またはBw54分子を認識することが明らかになった。
SCWに対する低応答者より得られたCD8^+T細胞は,リンパ球培養上清中の未知の液性因子存在下で単核球およびB細胞に対する細胞傷害活性をもつ特異な細胞である。今後,同細胞の低応答性への関与,液性因子の精製同定を行なっていく予定である。
