抗原ペプチドおよびT細胞レセプターとの結合に及ぼすHLA多型の影響に関する研究

Research Abstract	日本人集団において慢性関節リウマチ,インスリン依存型糖尿病などの自己免疫疾患の患者群で著明に増加しているHLA-DR4関連抗原を中心に、HLA-DR分子に結合しT細胞を活性化する非自己および自己由来の抗原ペプチドを同定し、これとHLA分子との結合において重要なペプチドおよびHLA分子上のアミノ酸残基を決定することを目的として研究を行った。DRB10406分子に結合した自己抗原ペプチドを認識するT細胞クローン3種類および溶連菌M蛋白に由来する21アミノ酸残基からなるペプチドを認識するT細胞クローンを一種類樹立した。前者は、DRB10402および0404分子をも認識することより、37Ser→Tyr,67Leu→Ileu,70Gln→Asp,71Arg→Glu,および74Glu→Alaへの置換は、自己抗原ペプチドの結合ならびにT細胞レセプターによる認識には関わっていないと推定された。いっぽう、DRB10401,0405および0410には全く反応しなかったことより、57Asp→Ser,71Arg→Lysおよび86Val→Glyの置換は抗原ペプチドの結合あるいはT細胞レセプターによる認識を不可能とすることが明かとなった。現在、DRB10406分子に結合する自己抗原ペプチドの精製ならびにアミノ酸配列の決定および溶連菌M蛋白に由来するペプチド上のT細胞エピトープの同定を実施しており、これらの情報をもとにDRB1*0406分子に結合するペプチドのモチーフを決定する予定である。
