| Research Abstract |
In this study we investigated the search of immunosuppressive substance on Cordyceps sinensis Sace. (Dong - Chong Xia Cao), the physiological mechanism and the chemical synthesis.
1. Isolation of ISP - I and the derivatives: Immunosuppressive agent, ISP - I was firstly isolated from the culture broth of Isaria sinclairii. The compound has also been obtained from C. sinensis of which the imperfect stage belongs to the same genus as Isaria. Mycelia is a high producing fungus of ISP - I. The culture broth gave two new immunosuppresive agents along with the three known ISP - I derivatives.
2. Immunosuppresive effect and the mechanism of ISP - I and the derivatives: The new immunosuppresive substances were examined for the mouse allogenic mixed lymphocyte reaction (MLR), the cytotoxity and the skin grafting. These results showed a similar effect to that of ISP - I. ISP - I indicated the specific inhibition against growth of IL - 2 dependent T - cells.
3. Synthesis of ISP - I: Synthesis of ISP - I was designed. The synthetic intermediate, cyclo (D - Val - 2 - amino - malonyl) bis - lactam dimethyl ether, has been derived from D - valine.
