Research Abstract

本年度は主としてheat shock proteins(hsp)に対する抗体を作製することに重点をおいた。即ち多くのhspはATPーbinding protein(ABP)である性質を利用し、HeLa細胞lysatesよりATPーagaroseを用いてABPを精製し、これを免疫原として単クロ-ン抗体(mAb)を開発した。その結果、70KDa hspに反応するいくつかのmabを得た(N7T2,N7T3など)。これらは、bovine脳より精製した70hspとも特異的に反応した。また、ラット70hspのcDNAをpET発現ベクタ-に組みこむことにより得たラット70hspリコンビナント蛋白とも反応することが確認されつつある。現在、これらのmabの70hspの各subーfamilyとの反応性を解析している。いずれにしても、これらのmAbは今後、hspの細胞表面発現機序や、T細胞との反応機序の解析に有効な手段を提出してくれるものと思われる。一方、我々は既にラット線維芽細胞WFBにHーras癌遺伝子を移入し、形質転換された際に発現する腫瘍抗原(TAAs)のひとつが実は70hspであり、しかも細胞表面に発現することを報告した。そこで次に、この抗原は70hspのどのsubfamilyに属するかを解析した。その結果、この抗原は70KDa heat shock cognate protein(hsc)であることが判明した。次に、TAAであるこの70hscが宿主免疫系と実際反応するか否かを検討した。WKAラットにBCGを注射し、末血や脾臓からCD4(ー)、CD8(ー)T細胞(DNT)をとり、これが70hscを構成的に発現する形質転換株W31に障害性に働くかどうかをみた。その結果、DNTはW31に明らかに障害性を示した。しかも、この反応性が70hscに対するmAbで阻止された。即ち、DNTが70hscをリガンドとして認識している可能性が示唆された。現在、DNTの特徴や、70hscが70hsc+抗原ペプチド、即ちpresenting moleculeとして働いている可能性も考え、ERでペプチドとの会合を阻止するbrefeldin Aなどを用いて研究中である。
