Research Abstract

前年度までにin situハイブリダイゼーションに適した標識プローブをDNA合成器で自動的に作成するシステムを確立した。本年度では、ビオチン標識の導入比を検討したところ、30merのオリゴDNAの3^′末端に10個のビオチンを組込むのが最適であることが判明した。ビオチンの導入比を下げると理論通りに検出感度が低下した。一方、導入比率を上げ15〜20にビオチン分子数を増加すると、やはり感度が低下した。この理由は明かではないが、核酸電気泳動法を用いた検討の結果、おそらく高ビオチン導入によりプローブ全体が疎水性になり、いかも様々な割合で重合を起こすことが認められた。さらに、ビオチンの導入部位として5^′末端およびプローブの配列中に組込んだ場合について、in situハイブリダイゼーションで詳細に検討した。5^′末端の組込みでは3^°末端への組込みとはほぼ同程度の検出感度が得られた。しかし、プローブ配列中への組込みは、たとえ1個のビオチン分子ですら、検出感度をほとんど0にした。これはハイブリダイゼーションの阻害によるものと考えられた。3^′および5^°末端のそれぞれに標識をした場合、同程度の感度が得られるという事実をもとに、両末端に10分子づつ計20分子のビオチンを標識したプローブについても検討した。確かに検出感度の向上を認められたが、その改善比は10〜20%程度と低く、しかもプローブを溶液中で重合しやすいことが判った。このため、経済的にみても、3^′ないし5^′末端のすづれかに10分子程度のビオチンを導入するのが最適であることが確立された。以上の実験はバゾプレシンmRNA検出を指標として行ったものであるが、まだ信じるに足る報告がみられないコリンアセチル基転移酵素について本法の有効性を証明しようと試みている。予備実験では実用化の可能性が大であるという感触が得られた。
