Research Abstract	平成3年度の研究により、フェニルセファロ-ス、DEAE-セファデックス、サクシニル-LDLセファロ-ス、ハイドロキシルアパタイトクロマトグラフィ-によるヒト血清からのCETP製精に成功した。精製CETPは分子量約68,000のほぼ単一バンドであり、これを抗原としてマウスを免疫し、ミエロ-マ細胞とのハイブリダイゼ-ションにより、ELISAにて陽性のクロ-ンを約10個得た。現在、Western blottingによって強い陽性反応を示すクロ-ンを選択中で、これにより抗原との反応性を検討した後、酵素免疫法を確立する予定である。また、同じ抗原を用いてヤギを免疫し、多クロ-ン抗体を得ることも試みている。本法をキット化するには多量のCETPの標準抗原が必要であるが、これを得るのに通常の精製法では多くのステップを要し、収率も十分ではない。そこで、我々は昆虫由来のBaculovirus expression systemを用いて、ヒトCETPのfull length cDNAをpVL1392expression vectorに組み込み、これをpolyhedrin promotorを有するACNPVとのhomologous recombinationにより、polyhedrinを細胞内に有しないプラ-クを選択することにより、CETP遺伝子を発現するクロ-ンをスクリ-ニングした。さらに、培養液中へCETP活性を分泌するクロ-ンを発見し、このクロ-ンの培養液中からCETPを精製せんと現在試みている。他方、CETP阻害因子の精製も進んでおり、阻害因子活性を有する分画を得ているが、活性は必ずしも高くなく、他の蛋白の混在もあり、さらに純粋な標品を得るべく検討を進めている。以上の研究過程において、血清からCETP及びその阻害蛋白を分離精製する際の第一段階の超遠心分離のため、Beckman超遠心機とスウィングロ-タ-を購入して使用した。
