Research Abstract

We have demonstrated that hepatocyte growth factor (HGF) is secreted from nonparenchymal liver cells and that a large amount of HGF is sequestered in the extracellular matrix(ECM) in Disse's space in the normal adult rat liver.
We also observed in this study that substances secreted from the primary cultured hepatocytes could release HGF from HGF-bound ECM.Analysis of the activating mechanism of latent HGF sequestered in ECM shows that two different substances, a proactivator of latent HGF (proactHGF) and an activating factor of proactHGF, participate in the activating latent HGF.A large amount of proactHGF is sequestered in ECM in normal rat liver.
These results indicate th epossibility that a cascade mechanism, first release an activating factor of proactHGF from hepatocytes after liver injury folowing activation(release) of latent HGF sequestered in ECM between the layr of hepatocyes and enthothelial cells forming sinusoid.
Thus, we postulate that a physiologically relevant protease, an activating factor of proactHGF, secreted from the partially injured hepatocytes has a key in the reparing mechanism after liver injury.
