神経情報伝達におけるカルシニュ-リンサブタイプの役割に関する研究

| Research Abstract |
カルシニュ-リンは中枢神経系に豊富に存在するタンパク質脱燐酸化酵素で、その活性はカルシウム/カルモジュリンによって調節されている。カルシニュ-リンはNMDA受容体、電位依存性カルシウムチャネル等の様々な機能蛋白の脱燐酸化を介して神経情報化を介して神経情報伝達において重要な役割を果たしていると考えられているが、その詳細は不明である。研究代表者らは遺伝子クロ-ニングの手法により、カルシニュ-リンの全一次構造を明かにし、さらにカルシニュ-リンには少なくとも2種類の異なる遺伝子によってコ-ドされるサブタイプが存在することを明らかにしてきた。本年度は以下のような成果をあげることができた。
1.合成オリゴヌクレオチドを用いたin situ hybridization法、及び大腸菌発現タンパク質を抗原として抗体を作製し、免疫組織化学的手法により、カルシニュ-リン各サブタイプのラット脳内における分布を調べ、カルシニュ-リンAα、Aβの2つのサブタイプが異なる分布を示すことを明らかにした。
2.精巣特異的に発現するカルシニュ-リン類似蛋白質のcDNAクロ-ニングに成功した。この分子はおそらく、精子のカルシウム依存的な運動性に関係するものと思われる。
3.ほ乳類でのカルシニュ-リンの機能を明らかにするためのモデル系として有用な酵母において、カルシニュ-リンBに相当する蛋白質のcDNAクロ-ニングに成功した。
4.カルシニュ-リンの活性化機構について、カルモジュリンアンタゴニストを用いて解析した。
現在は、各サブタイプの大量発現系を確立し、活性化機構をさらに明らかにすること及び、特異的阻害葉の開発を目的として研究を進めている。
