Bone Gla Proteinの前駆体の骨代謝における意義

Research Abstract	骨に特異的な蛋白質であるosteocalcin(以下、oc)はすぐれた骨代謝マーカーであるとされ、血清中ocの測定は臨床的に広く行われている。しかしながら骨代謝におけるocの真の役割は不明であり、この点を明らかにすベく、ocの前駆体pro osteocalcin(以下、pro oc)に着目し、この前駆体にのみ反応し、成熟蛋白質であるocには反応しない抗体を作製した。これを用いて骨腫瘍及び種々の骨リモデリングの状態の混在する大腿骨顕壊死症の摘出骨頭の免疫染色を試みた。この結果、骨腫において腫瘍細胞の細胞質内に強い抗pro ocの抗原性の存在を認め、既に報告したオステオカルシンの免疫染色以上に骨腫瘍の鑑別診断に有効性があるものと考えられた。また、ヒト骨肉腫細胞様を用いてWestern blotting analysisを行い、作製した抗pro oc抗体が細胞内pro ocのみ認識し、ocとは反応しない事を確認した。骨頭壊死症においてもpro oc免疫染色は、骨細胞の生死を鈍敏に検出し得ることから、pro ocは骨リモデリングを研究する上で有力な方法と言える。血清中pro oc抗体免疫交差反応性蛋白質については現在のところ、pro peptideとして小児にのみ存在するという見解と血清中には存在し得ないという報告がみられ、議論の一致をみていない。これは酵素抗体法による高感度、低特異性とradioimmuhoassayによる低感度、高特異性の矛循によるものと考えられ、現在両者の測定法を比較検討中である。
