| Research Abstract |
1.食用カエルの中腎組織のcDNAライブラリ-を作成し、ウシP450(11β)をプロ-ブに用いてプラ-クハイブルダイゼ-ション法でスクリ-ニングした。その結果、5個のポジティブクロ-ンが得られた。これらのクロ-ンのDNA塩基配列の決定を現在行っている。ウシ、ラット、ヒトのP450(11β)cDNAの塩基配列との類似性から、5個のクロ-ンはすべてP450(11β)をコ-ドしていることが予想された。
2.完全長のcDNAを持つと思われるクロ-ンを発現ベクタ-に組み込み、このものを培養COS細胞にエレクトロポ-レイション法で導入して発現させるため、現在予備実験を行っている。
3.ラットのP450(11β)とP450(11β,aldo)の各cDNAについて、制限酵素により切断ーライゲ-ション法によって、キメラcDNAを作成した。このものをCOS細胞中で発現させて、11、18、19水酸化活性とアルドステロン合成活性を担うタンパク示の領域の向定を行った。その結果、NH_2末端から約3分の2付近にあるオゾル(人名)の領域およびP450camのIーヘリックスに相当する領域が、酵素活性の特異性を決定するのに重要であることが分かった。
