| Research Abstract |
平成3年度の研究においてヒトサプレッサ-T細胞の作用機序について以下の新知見が得られた。
(1)サプレッサ-T細胞とB細胞をメンブレン・フィルタ-で仕切って接触を断つことによって,サプレッサ-機能が消失したことより,サプレッサ-T細胞はB細胞と直接接触することにより、B細胞の機能を抑制することが明かになった。
(2)抗LFAー1抗体(抗CD11a,抗CD18)及び抗ICAMー1抗体(抗CD54)の両者を培養系に加えることにより,サプレッサ-T細胞によるB細胞機能の抑制が大副に阻害された。しかし,他の抗体では,はっきりした阻害はみられなかった。これにより,サプレッサ-T細胞のB細胞機能抑制にはLFAー1/ICAMー1interactionが重要な働きをしていることが明かとなった。T細胞・B細胞をこれらの抗体で前処理したところ,T細胞上のLFAー1およびB細胞上のICAMー1をブロックした時にのみサプレッサ-機能が消失した。従って,ヒトB細胞上のICAMー1とサプレッサ-T細胞上のLFAー1との間の相互作用が重要であることが示唆された。
(1),(2)の知見は,サプレッサ-T細胞の機能発現メカニズムの解析に対して,画期的な貢献をなすものであり,この点からみて本研究は順調に進展していると考えられる。
