血管内皮細胞膜上での血液凝固調節因子トロンボモジュリンの機能と発現調節機構の解析

| Research Abstract |
内皮細胞の膜表面に発現しているトロンボモジュリン(TM)はトロンビンと複合体を形成し、トロンビンが持つフィブリン形成能、血小板凝集能、そして血液凝固VおよびVIII因子に対する活性化能などの血液凝固を促進する機能を阻害し、加えて、その複合体にはプロテインC活性化能が誘導され、トロンビンの産生をも抑制する。すなわちTMはトロンビンの血液凝固能を抗凝固能に変えてしまう血液凝固抑制因子である。TMの上記のような作用および役割は純化精製したTM標品を用いた研究から導かれた示唆であり、実際の内皮細胞上で上記の様な作用および役割を果しているか、否かは明かでなく推測の域を出ていなかった。
当該年の研究では1.培養ヒト血管内皮細胞上のTMはトロンビンと複合体を形成し、プロテインC活性化能を示すことを明かにしたのに加え、2.内皮細胞膜上でのTMのプロテインC活性化能は純化精製されたTMより1分子当りに換算して約10倍も高いこと、すなわち内皮細胞膜上のTMは精製TMで考えられていたより強い抗凝固機能を持つことを明かにした。またTMの発現調節の研究では昨年度の研究成果に加え、レチノイン酸がTNF処理で低下したTM産生を正常レベルまで回復させることを明かにし、悪性腫瘍時や炎症時に発症する血栓症を防ぐ糸口をレチノイン酸が切り開く可能性を提示した。
