| Research Abstract |
近年、生体防御反応、癌免疫において重要な役割を有すると考えられているインターロイキン1(IL1)にたいするナチュラルアンタゴニスト(IL1Ra)の在存が明らかになった。本研究では、IL1Raの作用機序の解明、担癌状態におけるIL1Ra産生の証明ならびに担癌にともなう免疫不全症、悪疫質における内因性IL1Raの病態生理作用を明らかにすることを目的とする。本年度の主な成果として1)マウスIL1Raを遺伝子組み替え法にて大腸菌にて大量発現し、完全精製することに成功した。精製IL1Raはマウス/ヒトIL1RにIL1と同等の親和性を有し、IL1の生物活性を特異的に抑制することが確認された。2)精製IL1Raをウサギに免疫することによりIL1Raにたいする特異抗体を作製した。この抗体を用いてマウスIL1RaにたいするELISAによる測定法を確立した。3)癌悪疫質発生マウス大腸癌細胞株colon26(clone-20)のinvivoにおけるサイトカイン、サイトカインナチュナルアンタゴニスト産生をnorthemblotting法にて検索した結果、癌組織浸織マクロファージ由来IL1により刺激を受けた癌細胞によりIL6が過剰に且つ持続的に産生され癌悪疫質をもたりしていることが判明した。一方、癌悪疫質非発生colon26(clone-5)においては、癌組織においてIL1の他、同時に大量のIL1Raが産生されており、結果的にIL6が全く新生されないことが判明した。colon26の実験結果は、IL1Raが癌組織において実際に産生されていることを証明し、さらに、IL1Raがサイトカインネットワークを調節する重要な内因性制御分子であることを示した世界で最初の仕事である。今後、種々の実験腫瘍、ヒト癌組織におけるIL1Ra、サイトカインの発現検索をするとともに担癌宿主におけるサイトカインネットワーク分子機構を生体工学、遺伝子導入法、単クローン性抗体等を駆使して解明する。
