| Research Abstract |
北村はc-kitレセプター・タイロシシカイネース9カイネース領域に小さな欠失を持つWs/Wsラットを用いて、好塩基顆粒を持つ3種類の細胞、すなわち結合組織型マスト細胞、粘膜型マスト細胞、好塩基球の分化に、c-kitレセプターを介したシグナル伝達がどの程度関与しているかしらべた。皮膚における結合組織型マスト細胞の分化には、c-kitレセプターを介したシグナル伝達が必須であるが、粘膜型マスト細胞の分化には、c-kitレセプターのリガンドであるstemcellfactor(SCF)とインターロイキン3(IL-3)の両方が必要であり、好塩基球の分化にはIL-3のみで十分であることがわかった。また日本で発見されたものも含めて、数種のマウスのc-kit突然変異をしらべ、おのおのの突然変異遺伝子の分子レベルの異常と、その突然変異遺伝子をホモあるいはヘテロに持つマウスの症状をくらべ、両者が整合することを示した。西川は造血組織以外で幹細胞を持つ睾丸や上皮組織と、造血組織を比較考察するため、"幹細胞システム"の中でc-kitレセプターとSCFに依存する組織を西川が作製した抗c-kitレセプター抗体を用いてしらべ、造血幹細胞と睾丸の精原細胞は、c-kitレセプターとSCF12恒常的に依存しているのに、卵母細胞やメラノサイトはある一定の時期をかぎってc-kitレセプターとSCFに依存することを示した。岡山は造血組織に特異的な細胞外マトリックス(ECM)の化学的組成を明らかにする目的で、ウサギ骨髄中のプロテオグリカンを分析したところ、他の組織のECMには存在しないタイプのプロテオグリカンをみつけた。そのプロテオグリカンは純粋なコンドロイチン6硫酸を糖側鎖とする分子量の比較的小さなものであることがわかった。仁保,三浦,仲野もひきつづき各自の研究課題について着実な進展を示した。
