Research Abstract	多能性造血幹細胞への遺伝子の導入は、基礎的には導入された幹細胞に於ける機能発現状態の解析、臨床的には将来的な遺伝子治療の面からの機能発現の制禦に関わる幹細胞研究の一課題を構成している。本研究は、標題の如く多能性幹細胞(CFU-S)への遺伝子導入とその機能発現に関する研究を目的とし、具体的には:1.mycの挿入された多能性幹細胞のCell Kinetics(理研:相澤慎一博士との共同研究)および、2.造血刺激因子受容体遺伝子導入マウスの作製およびこれによる幹細胞系の樹立(医科研:新井賢一教授との共同研究)のふたつの研究の重点をおいて進めることを計画していた。この期間中にこれら双方の課題で進展した事柄はつぎのような諸点である:1.mycを挿入したマウス骨髄細胞における造血幹細胞(CFO-S)ではトリチウム・チミジンの自殺試験によってS期分画の増加があり、しかしながらその子孫の分画では変異のみられないことからmycの幹細胞分画にはS期の拡大と併せてapoptosisによる調節が働くことが想定された。bc1-2などの発現検索をすすめたが、配間中に結論を得られなかった。研究の概略を国際実験血液学会で発表し、また国際誌Bxpt1Hematolに公表した。2.造血刺激因子受容体遺伝子導入マウスの作製の計画ではIL-3の受容体サブユニットのα鎖及びβ鎖の双方に対する「ト」マウスを樹立した。(遺伝子構築については既に発表の通り)。これらのマウスでの発現はβ鎖マウスについてのみ行った。2日間のIL-3存在下での液体培養の後、照射マウスで脾コロニー・アッセイを行うSpivak-Sasaki法での検索結果によれば、このβ鎖マウスでの脾コロニーの反応性は低濃度領域での立ち上がりが高感受性で、高濃度域では対照と差の見られない特異なパターンを示した。これはβ鎖にシグナルが入りα鎖はlimiting factorとなっているとのinvitroでの結果と矛盾ないが、α鎖遺伝子導入マウスの掛け合わせ実験でどのようになるか、今後の検討が注目されるものである。現在、両系統のマウスを交配中である。研究進展の概略を「病態生理」誌に概説した。
