| Research Abstract |
これまでの研究計画においてB細胞抗原受容体は、IgM分子とこれに結合するMB-1/B29を核とする複合体構造をしていることを示してきた。またMB-1分子の細胞内ドメインが抗原の刺激を細胞内、核内へと伝達する機能分子であることが明らかにされてきた。更に、この複合体からのシグナル伝達分子を明らかにすることを目的とし、本年度は、MB-1分子に結合するリン酸化タンパクp52を見出し、この分子のcDNAクローニングを行いタンパク構造、アミノ酸シークエンスの解析に踏み込んだ。p52はPMAによるPKC活性化によってそのリン酸化が誘導され、IgM、MB-1タンパクとコンプレックスとして共沈してくる。この分子に対する特異的なモノクローナル抗体(19-14)を作成し、タンパク性質、発現、細胞内でのチロシンリン酸化酵素との関連を解析した。λgt11-ベクターにタンパク発現させるcDNAライブラリーを作成し、19-14抗体によるスクリーニングを行った。その結果、α4cDNAクローンを単離し、この遺伝子のコードするタンパクをグルタチオンs-トランスフェラーゼとの融合タンパクとして作らせ、免疫学的に同一の抗原性を有することを示した。α4遺伝子のシークエンスを行い、推定アミノ酸解析を決定した。コンピューターによるホモロジー検索を行ったところ、これまでに報告されていない新しい遺伝子であることが確認された。
