| Research Abstract |
1)8q23-q24領域特異的microcloneからsingle-copy配列だけを集めprobeとしたヒトcomid libraryのscreeningで20個の陽性cosmidsを取得した。
2)それら及び既に8q遠位1/3にmapされているcosmidを用いて、TRPSI型患者とII型患者のmappingをFISH法により行った。両者共1copyを示したのは2個で、前者で2copy後者で1copyの可能性を示したのはなかった。
3)一部は2色FISH法により、cosmidsの配列順序をcen-1185-1148-1435-1512-553-1122-1173-telのように決定した。
4)1のcomidを用い、t(8;13)転座外骨腫患者DNAにおけるPFGE解析を行った。1cloneは患者において800kbのBsshII-aberrant fragmentを検出したので、これは外骨腫患者の転座切断点から600kb以内に位置すると考えられる。このcloneに対応するYAC-subclones中で切断点を検出するものを同定し、更に切断点そのものを含むDNA断片を追う予定である。
5)ヒトBMP1-cDNA由来のPCR産物を用いて釣り上げたcomid cloneは、その制限酵素地図および一部の塩基配列からBMP1を包含していたが、それはFISHにより8p21にマップされた。この結果は、ヒトBMP1座は8p21であり外骨腫遺伝子ではないことを示す。
