Research Abstract	(a)ハービマイシンの作用に関する研究:[^<14>C]ハービマイシンを調整し、v-Src蛋白のキナーゼ領域への結合を証明すると共に、結合によりチロシンキナーゼが失活する機構(ATPの結合阻止)を明らかにした。また、ヒトBCR/ABL遺伝子を導入したマウス骨随細胞と各種ヒト白血病細胞のハービマイシンに対する感受性の比較から、BCR/ABLチロシンキナーゼが抗がんの標的になることを示した。ハービマイシンはp210^<bcr/abl>チロシンキナーゼをv-Ablやv-Srcチロシンキナーゼをより強く抑制するが、EGFレセプターのチロシンキナーゼは阻害しないことから、ハービマイシン結合ドメイン構造の相違が考えられた。 (b)新規がん遺伝子機能抑制物質の探索研究:増殖因子要求性を指標とした検索系を確立し微生物代謝産物中に探索を行ない、Src,Ablトランスフォーマントで活性を示した新規物質YU-6247(仮名、アンスラキノン構造)を得た。本物質はSrcチロシンキナーゼをin vitroで阻害したので、新規チロシンキナーゼ阻害剤と結論された。 (c)プロテインキナーゼ抑制物質の新しい活性評価系:PKA、PKC、PTK、CaMKIIIの4種類のプロテインキナーゼ活性を同時に検出できる系を確立した。市販の阻害剤の作用の検討から、この系が選択性の高いプロテインキナーゼ阻害剤の効率的なスクリーニング系として有用である可能性が示された。
