| Research Abstract |
本重点領域研究の最終年度である本年度も、ポリオウイルス、インフルエンザウイルスおよびハラミクソウイルスを中心に研究を推進した。以下に主な研究成果を示す。
(1)ポリオウイルス受容体のマウスホモログがホモフィリックな結合様式を持つ細胞間接着分子であることを証明した(野本)。
(2)血流中のウイルスのポリオウイルス感受性トランスジェニックマウス体内での組織分布はヒトポリオウイルス受容体の発現に依存していることを証明した(野本)。
(3)ポリオウイルスのプラス鎖RNA合成開始に関わる新しいウイルスゲノム領域の同定に成功した(野本)。
(4)ポリオウイルス蛋白質合成開始に関わる複合体に含まれるPTBとの結合性を示す蛋白質の一つは86kDaであることを示唆するデータが得られた(豊田)。
(5)PTB結合蛋白質のcDNAを酵母twohybrid systemにより分離した(野本)。
(6)インフルエンザウイルスのRNA合成酵素の各サブユニットを大腸菌で作る系が確立し、各サブユニットを再構成することに成功した(石浜)。
(7)さらに、この実験系を利用し、各サブユニット間の結合に関わる部位、およびRNAに結合している部位が同定された(石浜)。
(8)インフルエンザウイルスRNA合成におけるRAF(RNA polymerase activation factor) およびRRF(RNA polymerase regulation factor)の精製が進行した(永山)。
(9)パラミクソウイルスNDVのHN蛋白のmRNAがインターフェロン処理により特異的に阻害を受けることが明らかとなり、各mRNA合成がVRNA3'末端から順番に起こるという定説に疑問を投げかけた(永井)。
(10)パラミクソウイルスによる細胞融合に関与する細胞側分子として、VLA3およびCD98が同定された(伊藤)。
(11)パラミクソウイルスRNA合成に負に働く宿主細胞側の宿主分子(100-150kDa)がラット肝に存在することを見出した(水本)。
以上のように、ウイルス複製に関するウイルス側および宿主側分子群の分離・同定およびその構造と機能の研究が順調に進んだ。
