Research Abstract	パラインフルエンザウイルスSV41のP遺伝子に見い出されたRNA Editingを解析するために、このP遺伝子の上流に近縁のパラインフルエンザウイルスPIV2のリーダー配列(およびR1配列)、下流にPIV2のトレイラー配列を組み込んだcDNAを構築した。このcDNAをブルースクリプトIIベクターのT3プロモーターの下流に組み込みT3RNAポリメラーゼを用いてゲノムセンスRNA(vRNA)をランオフ合成した。この合成RNAをPIV2感染HeLa細胞に導入して20〜24時間後に細胞からRNAを分離精製し、リバースPCR法によりvRNAあるいはmRNAセンスRNA(CRNE)の解析を行った。その際果、1)R1配列を含むRNAの場合にはCRNAのみが検出され、PV41のP遺伝子がmRNAに転写されていることが示唆されたので、PCR物産をクローニングしてその塩基配列を調べたが、現在までのところEdited RNAは見つかっていない。2)R配列を含まないRNAの場合にはvRNAおよびcRNAの両センスのRNAが検出させ、RNAの複製が起こっていることが示された。なお、RNAを導入した細胞の上澄みを新たなHeLa細胞に接種したところ2代継代後の細胞内にもvRNAが検出されたので、P遺伝子のEncapsidationおよびPackagingが起こっていると考えられる。次にPIV2ゲノムをすべて被覆する6個のcDNAクローンをpUCL19ベクターに構築した。そこから膜融合遺伝子Fと膜融合調節遺伝子HNをpcDLS Rαベクターにそれぞれ組み換えてHeLa細胞で糖蛋白として発現させ、その機能(細胞融合能)が確認できた。さらにゲノムサイズのcDNAを講築し、ワクシニアベクターから細胞内発現させたT7RNAポリメラーゼで全構成蛋白を発現させてウイルス粒子を形成させるために、ブルースクリプトIIベクター系での発現用クローンを作製中である。
