Research Abstract

研究の目的:光合成の効率に影響を与える環境要因としては、温度、塩、無機イオン欠乏などが挙げられるが、これまでの研究により、上記の環境因子への適応に関する多数の遺伝子が単離された。本年度は、これらの遺伝子を他の植物・藻類・シアノバクテリアに導入し、さらに作成された形質転換植物の表現型を解析することを目的とした。
主要研究成果:(1)低温適応・耐性:不飽和化酵素の遺伝子を順次破壊した形質転換シアノバクテリアを用いて、膜脂質の不飽和化が光化学系II複合体の光失活からの回復を促進することを明らかにした。(2)塩耐性:塩耐性に関与するペタインの合成系の遺伝子(大腸菌由来)をシアノバクテリアに導入し、シアノバクテリアを塩耐性にすることができた。(3)乾燥による遺伝子発現:乾燥によって誘導されるシロイヌナズナの遺伝子rd29Aのプロモーター領域を解析し、乾燥によって直接制御を受けるシス領域とアブサイシン酸を介して制御を受けるシス領域を同定した。(4)シアノバクテリアからP・型ATPaseの遺伝子をクローニングした。その遺伝子産物はチラコイド膜に局在し、銅イオンの輸送に関与していた。P・型ATPaseの遺伝子をもう一種類クローニングしたが、その遺伝子産物はカルシウムの輸送に関与していた。
