Research Abstract

転写の包括制御は、転写装置の転写対象遺伝子選択特性の変換によっていることが示唆されている。転写制御の本質を理解することを終局の目標として、転写装置の分子解剖を行なうことが、現在の主要な研究課題となってきた。そのための研究推進方策を検討する目的で、本年度以下の活動を展開した。
1)シンポジウム「核酸を認識するタンパク質の構造」(平成4年11月2日・東京)、研究集会「RNAポリメラーゼ:構造とダイナミクス」(平成5年1月11日・三島)を開催し、転写装置の分子解剖に関する研究動向の調査分析を行った。
2)転写装置の分子解剖に関する研究の国内外の現状と動向を調査分析した。文献調査に加えて、直接面談調査の結果、当該分野では、世界15ケ国100件以上の国際共同研究が実施されていることが判明した。しかし、この領域の重点領域研究の指定がないため、いずれの研究室も経済的基盤は
3)転写をめぐる研究動向に関する専門家討論会(平成4年12月26日・名古屋)を開催し、今後の研究方向として、RNAポリメラーゼを核とする転写装置の分子遺伝学的構造ー機能相関分析、NMR・X線・電子顕微鏡を主武器とした構造分析、バイオインフォーマティクスによる構造機能不析と一分子ダイナミクス観測などによる動態分析の重要性を指摘した。
これらの調査研究の結果、研究推進のひとつの対策として、重点領域研究“転写装置の機能構造"を提案した。
