新しい肺保存液の開発とADF/hチオレドキシンのラジカルスカベンジャー効果

Research Abstract

肺保存に対するトレハロースの有効性:
ユーロコリンズ液(ECS)とグルコースに換え3.5重量%トレハロースの両者を用いて12時間保存肺移植を行なった。トレハロース群はユーロコリンズ群と比較し、術後、移植肺のPaO2が有意(p<0.05)にで良好であった(Surgery,accepted)。ついでtrehaloseを糖質源に、Gluconateを陰イオン源に、HESを膠質浸透圧源にした低K_+、高Na_+の細胞外組成の新しい肺保存液ET-K液を開発し、20時間冷所保存を行った。対照としたECS(±PgE1)群と比較し有意に(p<0.05)ET-K群が良好な肺機能を示した。続いて、48時間保存実験を行ったが、術後6時間の経過観察で、PaO2が100mmHg前後であった。現在、。燐酸バッファーを強力にする時、Gluconateを減量したところ保存能が悪化したので、構成要素の分析を行いながらET-K液のバッファー機能強化と陰イオン源の有効な組成を検討している。
ADF/hThioredoxinのラジカルスカベンジャー効果:
ADFThioredoxinは104個のアミノ酸からなる2つのSH基を有する還元物質であり、その作用にラジカルスカベンジャー活性がある。ラットのマッチドペアー法による肺再潅障害実験で30μg/gのADFを投与した。ADF投与群は非投与群に比較し有意に良好なPaO2を示した(p<0.05)。再灌流肺組織中のSOD、Gluthathion Peroxdase,脂質過酸化物の量は、ADF投与群で非投与群に比較し有意に低値を示した(p<0.05)。
FK506大量投与による免疫寛解誘導:
免疫抑制剤の検討ではFK506とCyAの併用群は各々の単独使用群より有意に良好な拒絶反応抑制効果を示したので、FK506大量投与による免疫寛容誘導の実験を行い(トロント)、通常量の10倍を投与したところ、30日間免疫抑制が不要であった。
