平滑筋および血管内皮細胞のエンドセリン受容体のサブタイプと生理機能に関する研究

Research Abstract

従来、平滑筋にはET_A受容体があり、収縮に関与するが、内皮細胞にはET_B受容体があり、内皮依存性弛緩因子の放出に関与することが報告されてきた。本年度はこの点を確認すると共に、これら受容体のサブタイプ分けを試みた。その結果、静脈平滑筋において収縮に関与するET_B受容体が存在し、これはET_<B1>受容体とET_<B2>受容体に細分類されることを世界に先駆けて明らかにすることができた。さらに最近、我々はET_A受容体もまたET_<A1>受容体とET_<A2>受容体に細分類できることを見いだした。
さらに、内皮細胞は血管内腔表面だけではなく、血管各部位や心臓の弁にも存在する。ウサギ大動脈弁のエンドセリン受容体について検討したところ、ここにはET_A受容体のみが存在しており、従来内皮細胞にはET_B受容体のみが存在するという結果とは異なる成績が得られた。またこの受容体を介する細胞内Ca濃度上昇の機序についても明らかにすることが出来た。
その他、平滑筋のエンドセリン収縮の機序、エンドセリン収縮に対する弛緩薬の影響などについても検討を行った。
詳細は添付した論文集に譲るが、これらの発見がエンドセリンの生理的、病態生理的役割の解明に寄与するものと期待される。
