Research Abstract

Hematopoiesis is regulated by microenviornment created by stromal cells in hematopoietic organs. To understand the regulation of growth and differentioation of hematopoietic stem cells and their progenitor cells by stromal cells, we tried to reconstruct in vitro hematopoietic microenvironment by using the established stromal cells. We established many stromal cell lines from fetal liver, spleen and bone marrow of mice and characterized their ability to support hematopoiesis in vitro. We demonstrated that the stromal cell lines established from the erythropoietic organs supported in vitro erythropoiesis and their supporting ability is depend on c-Kit (a recepter type proto-oncogene product)-S1(a ligand for c-Kit) interaction, VCAM-1-VLA4 adhesion and erythropoietin.
Bone marrow stromal cells supported selectively the growth and differentiation of erythroid, granulocytes and macrophages. Furthermore, we obtained the stromal cell lines that supported in vitro maintenance of hematopoietic stem cells. Using these stromal cell lines, it is possible to identify the molecules on the stromal cells essential for hemtopoiesis.
