Research Abstract

1.熱帯熱マラリア原虫スポロゾイト表面タンパク質(CSP)遺伝子の多型:CSP遺伝子の多型をタイで分離した18株について調べた。CSPの主要T細胞エピトープであるTh2R,Th3R,CTL領域に変異が集中していたが、CS・T3では変異が認められなかった。主要B細胞エピトープであるテトラペプチド反復領域では、点突然変異、反復単位の欠出・挿入、遺伝子内組換え、という三つの機構が多型の形成機構であると示唆された。また、T細胞エピトープとテトラペプチド反復部位との間では変異の遺伝的連関性はなかった。
2.熱帯熱マラリア原虫メロゾイト表面抗原(MSPI)遺伝子の多型:タイ分離株についてMSP1遺伝子の多型をブロック1-4、ブロック13-17領域で検討した。3′末端側ではMSP1の代表対立遺伝子であるMAD型とK1型の遺伝子内組換えは認められなかった。変異塩基数は5′側の保存領域と比べて約1/2と少なく、18個あった変異はすべてdimorphicで、非同義語置換になっていた。C末端側の19kDポリペプチド部分に存在する12個のシステイン残基は完全に保存されていた。一方、19kD切断部位のオリゴペプチドに反応するモノクローナル抗体を作成し、そのエピトープを分子マッピングして同定した。また、MSP1の分子疫学的解析を行うために、血液塗抹標本から原虫DNAを分離するという技術を開発した。
3.MSP1T細胞エピトープ部位の決定:MSP1特異的なT細胞のクローンを3人の日本人から確立した。MSP1のブロック1-6のうち、CD4^+・8^-クローンが反応したのは変異領域であるM6ブロックのポリペプチドのみであった。13-16残基の合成オリゴペプチドを用いた抗原特異的増殖で見ると、それぞれのT細胞クローンは5種類以上の異なるペプチドを認識し、その反応性にはMHCの拘束性の存在が示唆された。
