成人T細胞白血病ウイルスによる関節炎発症機構の解明

Research Abstract

我々はHTLV-IのpX領域を導入したトランスジェニックマウスを作製し、関節リウマチに似た慢性関節炎が起こることを示した(Science,253,1026,1991)。本研究は、このトランスジェニックマウスを用いて、関節炎の発症機構を解析すると共に、ヒト関節リウマチモデルとしての有用性を検討したものである。
関節では、滑膜細胞の増殖と絨毛の過形成、炎症性細胞の浸潤と、骨・軟骨の破壊、パンヌスの形成が認められ(Rheum.Arth.36,1612,1993)、リウマチ因子の他、II型コラーゲンや熱ショック蛋白に対する自己抗体が検出されるなど、病態が関節リウマチに似ていた(投稿中)。このほか、免疫グロブリンの糖鎖は末端ガラクトースを欠損しているものが多く(BBRC,192,1004,1993)、関節ではIL-1α、IL-2β、IL-2、IL-6、TNF-α、IFN-γなどの発現が亢進していた(投稿中)。このようにこの関節炎の病態は、ヒトの関節リウマチのものときわめてよく似ていることがわかった。このことは、HTLV-Iがヒトの関節リウマチにも関与している可能性を示唆すると共に、このマウスが関節炎のモデルとして有用であることを示している。
関節炎の発症機構については、関節局所での炎症性サイトカインの異常発現と自己免疫の果たす役割が注目される。関節炎の発症率に対する遺伝的な影響を検討したところでは、系統により大きな差があり、関節炎の発症に免疫反応が関与していることが示唆されている(論文作成中)。このようにこのマウスは、関節リウマチの発症機構だけでなく、自己免疫の発症機構やサイトカインと免疫異常との関連など多くの重要な問題を解析するためのよい系となることが期待できる。今後この系を用いて、レトロウイルスと宿主の相互作用をより詳細に解析する予定である。
