| Research Abstract |
実験動物(ネコ)及びヒト手術肺の気道から粘膜下腺を単離して検索し、次のような所見を得たが、その際、実験動物とヒトでは明瞭な差異はみられなかった。
1)単離粘膜下腺を酵素処理して得られた腺房細胞にパッチクランプ法を応用し、Whole cellの状態でアセチルコリン、サブスタンスp及びブラディキニンに反応して外向きのK^+チャンネルと内向きの電流のCl-チャンネルが生じた。これらの電流応答は細胞内IP_3投与によるものと類似していた。しかし、β-刺激剤、TBMX及び細胞内サイクックAMP投によっては有意の電流応答がみられなかった。カフェイン及びcyclicADPribov(CADPR)では主にK^+電流が認められた。更に、粘膜下腺からの腺房細胞でのパッチクランプ法でLarge conductanceの,K_+チャンネル(160ps)が見い出され、ヒト肺からの粘膜下腺細胞の培養細胞の管腔側膜に3種類のCa^<++>依存性Cl^-チャンネル(3、20及び40ps)が認められた。
2)[Ca^<++>]iの画像解析では[Ca^<++>]iの上昇の局在性がみられ管腔側に初期の上昇がみられた。IP_3受容体に対するモロクロナール抗体を用いた組織学的検索でIP_3受容体が管腔側の細胞内膜成分に限局していることを認めた。
3)単離粘膜下腺からの膜成分を用いたレセプターアッセイを用いて、ムスカリン受容体のサブタイプを検索し、M_3が主であることを認めた。
以上より、気道粘膜下腺細胞で受容体刺激によりIP_3が生成され、管腔側に局在するIP_3感受性Ca^<++>プールよりCa^<++>放出が生じてCl^-電流が生じ、K^+電流はCADPRによりカフェイン感受性のCa^<++>プールからのCa^<++>放出により生じることが推定された。
