Research Abstract |
In the first year of this project, we built up the surface plasmon microscope (SPM) and established the preparation method of specimen. The SPM was newly designed involving the computer controlled adjustment mechanism according to the experience of prototypes. This new design increased the system robustness and resulted rapid sample exchange. As to the specimen, silver was evaporated to the glass, initially. However, silver oxidization occurred and the silver membrane was not tightly attached to the glass. Then, multi layred membrane, chromium and gold, was employed to improve tightness. Polymer membrane which was spin coated on the metal layr, had some problems such as its uniformity and foam formation. These problems are fixed up with adjusting the temperature and atmosphere for after bake. In the last year, we have studied on two theme. First, we pursued the real time measurement of protein adsorption onto the surface of bio-compatible material (polyurethane) and metal electrode (chromium-gold). The albumin adsorption on the polyurethane surface indicated two phase in its time course. The initial rapid phase counted its time constant as 7 minute. This rapid phase was followed by the next slow phase, which time constant was 2 hours, approximately. Gammaglobulin firmly adsorbed not only on the polyurethane membrane but on the chromiumgold surface, in spite of inorganic material. Secondary, we evaluated the effect of surface electrostatic field on protein adsorption. The inhibition of protein adsorption was observed under electrostatic field, which was applied initially before the exposure of protein solution. However, after the completion of protein adsorption, the electrostatic field did not influence the thickness of protein layr. The study has been continued on micro fabricated electrode which applied horizontal electrostatic field within the surface. Also, the effect of AC electrostatic field is still in experiment.
